ABSTRACT

Abstract Vegetative aerial organs are considerably more exposed to environmental conditions and can reflect the specific adaptations of plants to their local environment. Aldama grandiflora species are known to be widely distributed in Brazil; therefore, individuals from different populations of this species are thought to be exposed to different abiotic and biotic conditions. Several anatomical studies conducted on Brazilian Aldama species have mainly focused on the qualitative anatomical characters or traits of these species, but not on their quantitative traits. In this study, we evaluated whether climate and soil conditions can change the morphometry among individuals of A. grandiflora collected from six sites in the Goiás State, Brazil, by assessing their anatomical characters. Further, soil sampling was performed, and climate data were collected from all the six sites. The analysis indicated few statistical differences among the populations evaluated, showing that A. grandiflora presented consistent leaf and stem anatomical characteristics. The small morpho-anatomical differences found among individuals of the different populations evaluated, reflected the soil conditions in which these populations were grown. Therefore, environmental factors have a significant influence on the morpho-anatomy of Aldama grandiflora.


Resumo Os órgãos vegetativos aéreos estão consideravelmente mais expostos às condições ambientais e podem refletir as adaptações específicas das plantas ao seu habitat. A espécie Aldama grandiflora é amplamente distribuída no Brasil e, dessa forma, indivíduos de diferentes populações podem estar expostos a diferentes condições ambientais. Vários estudos anatômicos realizados com espécies brasileiras do gênero Aldama têm abordado, principalmente, as características anatômicas qualitativas dessas espécies, mas não em suas características quantitativas. Neste estudo avaliamos se as condições climáticas e do solo podem alterar a morfometria entre os indivíduos de A. grandiflora coletados em seis populações do Estado de Goiás. Foram avaliados os caracteres anatômicos foliares e caulinares, além da amostragem do solo e coleta de dados climáticos, para os seis locais. A análise indicou algumas diferenças estatísticas entre as populações avaliadas, mostrando que A. grandiflora apresentou características anatômicas foliares e caulinares bastante consistentes. As pequenas diferenças morfo-anatômicas encontradas entre indivíduos das diferentes populações avaliadas, refletiram as condições do solo nos quais essas populações se desenvolveram. Assim sendo, fatores ambientais relacionados ao clima e condições do solo têm uma influência significativa sobre a morfo-anatomia de Aldama grandiflora.

ABSTRACT This study aims the anatomical description and chemical characterization of aerial parts of Clusia criuva Cambess., Clusiaceae in addition to the evaluation of the antioxidant activity of crude extracts, correlated to the flavonoid content. The morphological characterization was performed using traditional techniques of plant anatomy. For phytochemical studies, crude extracts were obtained by static maceration and analyzed by thin layer chromatography. The antioxidant activity and the flavonoids content were determined by colorimetric methods involving, respectively, 2,2-diphenyl-1-picrylhydrazyl free radical and aluminum chloride. C. criuva has uniseriate epidermis, paracytic stomata, hypostomatic leaves, cuticular flanges and cordiform vascular cylinder with accessory bundles. Chemical prospecting confirmed the abundant presence of terpenes and phenols in the extracts of leaves and of fruits. The methanolic extract of seeds showed the lowest EC50 value, but the methanolic extract of pericarps exhibited the highest maximum antioxidant activity. The results suggested a high percentage of flavonoids in the hexanic extract of pericarps, however, this could represent, in fact, the presence of benzophenones. Secretory ducts and the shape of the midrib are diagnostic for C. criuva. The antioxidant activity is not directly related to the flavonoids. The results indicate the importance of future studies with C. criuva chemical constituents.

ABSTRACT

BACKGROUND: The epicuticular waxy layer of plant leaves enhances the extreme environmental stress tolerance. However, the relationship between waxy layer and saline tolerance was not established well. The epicuticular waxy layer of rice (Oryza sativa L.) was studied under the NaHCO3 stresses. In addition, strong saline tolerance Puccinellia tenuiflora was chosen for comparative studies. RESULTS: Scanning electron microscope (SEM) images showed that there were significant changes in waxy morphologies of the rice epicuticular surfaces, while no remarkable changes in those of P. tenuiflora epicuticular surfaces. The NaHCO3-induced morphological changes of the rice epicuticular surfaces appeared as enlarged silica cells, swollen corns-shapes and leaked salt columns under high stress. Energy dispersive X-ray (EDX) spectroscopic profiles supported that the changes were caused by significant increment and localization of [Na(+)] and [Cl(-)] in the shoot. Atomic absorption spectra showed that [Na(+)]shoot/[Na(+)]root for P. tenuiflora maintained stable as the saline stress increased, but that for rice increased significantly. CONCLUSION: In rice, NaHCO3 stress induced localization and accumulation of [Na(+)] and [Cl(-)] appeared as the enlarged silica cells (MSC), the swollen corns (S-C), and the leaked columns (C), while no significant changes in P. tenuiflora.

ABSTRACT

Extraneous DNA interferes with PCR studies of endophytic fungi. A procedure was developed with which to evaluate the removal of extraneous DNA. Wheat (Triticum aestivum) leaves were sprayed with Saccharomyces cerevisiae and then subjected to physical and chemical surface treatments. The fungal ITS1 products were amplified from whole tissue DNA extractions. ANOVA was performed on the DNA bands representing S. cerevisiae on the agarose gel. Band profile comparisons using permutational multivariate ANOVA (PERMANOVA) and non-metric multidimensional scaling (NMDS) were performed on DGGE gel data, and band numbers were compared between treatments. Leaf surfaces were viewed under variable pressure scanning electron microscopy (VPSEM). Yeast band analysis of the agarose gel showed that there was no significant difference in the mean band DNA quantity after physical and chemical treatments, but they both differed significantly (p < 0.05) from the untreated control. PERMANOVA revealed a significant difference between all treatments (p < 0.05). The mean similarity matrix showed that the physical treatment results were more reproducible than those from the chemical treatment results. The NMDS showed that the physical treatment was the most consistent. VPSEM indicated that the physical treatment was the most effective treatment to remove surface microbes and debris. The use of molecular and microscopy methods for the post-treatment detection of yeast inoculated onto wheat leaf surfaces demonstrated the effectiveness of the surface treatment employed, and this can assist researchers in optimizing their surface sterilization techniques in DNA-based fungal endophyte studies.

ABSTRACT

We present the leaf anatomy of seven amphibious species of Aeschynomene L. (Papilionoideae, Leguminosae), interpreting their structures and ecological functions, and also, providing information on which their taxonomy can be based, especially of morphologically similar species. We evaluated Aeschynomene americana, A. ciliata, A. evenia, A. denticulata, A. fluminensis, A. rudis and A. sensitiva. The anatomy corroborates the separation of the series Americanae, Fluminenses, Indicae and Sensitivae, with the shape of the petiole, types of trichomes and quantity of vascular units in the petiole as main characteristics to delimit the species. The petiole shape varies from cylindric in A. americana, A. sensitiva and A. fluminensis, to triangular in A. evenia and quadrangular in A. rudis, A. denticulata and A. ciliata. We observed four types of trichomes: hydathode trichome, long conic trichome, short conic trichome and bulb-based trichome. The hydathode trichome was the most common, except for A. americana and A. fluminensis. Species with higher affinity with water share similar adaptive characteristics, including hydathode trichomes described for the first time for the genus. This article adds unseen descriptions for the genus and on the adaptation factors of the amphibious species.


Neste trabalho é apresentada a anatomia foliar de sete espécies anfíbias de Aeschynomene L., interpretando estruturas e suas funções ecológicas e fornecendo informações que subsidiam sua taxonomia, principalmente de espécies semelhantes morfologicamente. Foram avaliadas Aeschynomene americana, A. ciliata, A. evenia, A. denticulata, A. fluminensis, A. rudis e A. sensitiva, pertencentes ao grupo Papilionoideae - Leguminosae. A anatomia corrobora a separação das séries Americanae, Fluminenses, Indicae e Sensitivae, sendo o formato do pecíolo, tipos dos tricomas e quantidade de unidades vasculares no pecíolo as principais características para delimitação das espécies de Aeschynomene. O formato do pecíolo variou de cilíndrico para A. americana, A. sensitiva e A. fluminensis, triangular para A. evenia e quadrangular para A. rudis, A. denticulata e A. ciliata. Foram observados quatro tipos de tricomas, são eles: tricoma-hidatódio, tricoma cônico longo, tricoma de base bulbosa e cônico curto. O tricoma-hidatódio foi o mais comum, ausente somente em A. americana e A. fluminensis. Espécies com maior afinidade pela água compartilham características adaptativas similares, incluindo tricoma-hidatódio descrito pela primeira vez para o gênero. Este trabalho acrescenta descrições inéditas para o gênero e sobre fatores adaptativos das espécies anfíbias.

ABSTRACT

Alternaria sesami causes leaf spot disease in Sesamum orientale. Conidium germination, inoculation, penetration and colonization of the pathogen on the plant surfaces were studied using scanning electron microscopy. Electron microscopy analysis revealed multiple germ tubes from conidium that spread in all direction across the leaf surfaces. Penetration in the plant surface occured, directly through the epidermis or via stomata with or without the appressoria formation. Hyphal penetration continued through the substomata cavity and some of hyphal branches grew in the intercellular space of mesophyll tissue. Hyphal toxin, caused cell and cell wall damages. Changes in different biochemical parameters in the diseased sesame plants (both in wild and cultivar) were compared to control. Transmission electron microscopy showed structural changes in the chloroplast of diseased plants. Isozyme pattern and assays of different enzymes, namely catalase, acid phosphatase and peroxidase expressed varied level of activities. Meanwhile, esterase, polyphenol oxidase and superoxide dismutase in diseased plants showed remarkable levels compared to control. Due to the infection, chlorophyll content, carbohydrates and total soluble protein decreased whereas free amino acid, proline, phenols and disease-related proteins increased in the host plants. Differential SDS-PAGE band profiling of total soluble proteins were also observed in plants due to the infection.

ABSTRACT

The genus Zamia is morphologically and ecologically the most diverse of the order Cycadales. Throughout its history this genus has been restricted to the New World and is presently almost entirely restricted to the Neotropics. Unusual anatomical traits of the leaflets, such as the sunken stomata and thick cuticle, are common in this and related genera. The objective of this research was to study and compare the leaflet anatomy of Zamia acuminata and Z. pseudomonticola and establish possible phylogenetic relationships between the anatomical traits and the near relatives of these species. The leaf material was obtained from living plants and then processed for electron microscopy study. We found that both species are very similar to each other and to Z. fairchildiana, and that they share several unusual traits with other species of the genus, such as the parenchyma morphology, the spatial distribution of tissues between the veins and the stomata morphology. The main differences between these species were seen in their fiber clusters and in the abundance of trichome basal cells on the epidermis. The anatomical similarities between the three species could be the result of their close phylogenetic relationship and the divergences between them could be the result of recent speciation during the Pleistocene, resulting from geological changes in Southern Costa Rica.


Zamia es morfológica y ecológicamente el género más diverso del orden Cycadales. Este género siempre ha estado restringido a América, pero en la actualidad habita principalmente en la región neotropical. Características anatómicas inusuales en los foliolos como los estomas hundidos y las cutículas gruesas son comunes en Zamia y géneros afines. El objetivo de este trabajo consiste en comparar la anatomía de los foliolos de Zamia acuminata y Z. pseudomonticola y establecer posibles relaciones filogenéticas entre las características anatómicas y los parientes cercanos de esta especie. Las hojas de las especies seleccionadas fueron obtenidas de plantas vivas y luego procesadas para el estudio por microscopía electrónica. Ambas especies son muy similares entre sí y respecto a Z. fairchildiana y comparten varias características en común con otras especies del género como son la morfología del parénquima, de los estomas y la distribución espacial de tejidos alrededor de las haces vasculares. Las diferencias más notables entre especies se vieron a nivel de sus paquetes de fibras y en la abundancia de células basales de los tricomas en la epidermis. Las similitudes anatómicas entre estas tres especies pueden ser el resultado de su cercanía filogenética y las diferencias podrían ser el resultado de especiación durante el Pleistoceno, producto de los eventos geológicos y cambios sucedidos en el sur de Costa Rica en esa época.

Smilax L. in Brazil is represented by 32 taxa and it is a taxonomically difficult genus because the plants are dioecious and show wide phenotypic variation. The analysis and use of leaf anatomy characters is recognized as a frequently successful taxonomic method to distinguish between individual taxon, when floral material is absent or minute differences in flowers and foliage exist such as in Smilax. The aim of this study was to characterize the anatomical features of the aerial organs in Smilax syphilitica collected from the Atlantic Rainforest, in Santa Teresa-ES and the Smilax aff. syphilitica from the Amazon Rainforest, in Manaus, Brazil. For this, a total of three samples of Smilax were collected per site. Sample leaves and stems were fixed with FAA 50, embedded in historesin, sectioned on a rotary microtome, stained and mounted in synthetic resin. Additionally, histochemical tests were performed and cuticle ornamentation was analyzed with standard scanning electron microscopy. S. syphilitica and S. aff. syphilitica differed in cuticle ornamentation, epidermal cell arrangement and wall thickness, stomata type and orientation, calcium oxalate crystal type, and position of stem thorns. Leaf blades of S. syphilitica from the Amazon Rainforest have a network of rounded ridges on both sides, while in S. aff. syphilitica, these ridges are parallel and the spaces between them are filled with numerous membranous platelets. Viewed from the front, the epidermal cells of S. syphilitica have sinuous walls (even more pronounced in samples from the Amazon); while in S. aff. syphilitica, these cells are also sinuous but elongated in the cross-section of the blade and arranged in parallel. Stomata of S. syphilitica are paracytic, whereas in S. aff. syphilitica, are both paracytic and anisocytic, and their polar axes are directed towards the mid-vein. Calcium oxalate crystals in S. syphilitica are prisms, whereas in S. aff. syphilitica, crystal sand. Thorns occur in nodes and internodes in S. syphilitica but only in internodes in S. aff. syphilitica. These features have proven to be of diagnostic value and may support a separation into two species, but future studies are needed to confirm that S. aff. syphilitica is indeed a new taxon.


Smilax L. en Brasil está representado por 32 táxones y es un género difícil en lo que respecta a su taxonomía porque las plantas son diocas y presentan mucha variación fenotípica. El análisis y uso de los caracteres anatómicos de la hoja es reconocido frecuentemente como un exitoso método taxonómico para distinguir entre los táxones individuales cuando las flores están ausentes o hay diminutas diferencias. El objetivo de este estudio fue caracterizar los rasgos anatómicos de los órganos aéreos en Smilax syphilitica recolectados en el Bosque Atlántico en Santa Teresa- ES y de Smilax aff. syphilitica del Amazonas, Manaus, Brasil. Un total de tres muestras fueron recolectadas por sitio. Las muestras fueron fijadas en FAA 50, sumergidas en historesina, seccionadas en un micrótomo rotatorio, teñidas y montadas en resina sintética. Se hicieron pruebas histoquímicas y la ornamentación de la cutícula fue analizada con microscopía electrónica de barrido estándar. S. syphilitica y difirieron en la ornamentación de la cutícula, la disposición de células epidérmicas y el grosor de la pared, tipo y orientación de estomas, tipo de cristal de oxalato de calcio y la posición de las espinas del tallo. Estas características han probado ser de valor diagnóstico y pueden apoyar la separación de dos especies, pero se necesitan futuros estudios para confirmar que S. S. aff. syphiliticaaff. syphilitica es un nuevo taxon.

Cuphea aquipetala Cav. (Hierba del cancer) is used in Mexican traditional medicine. In the present work, its most frequent medicinal applications were determined through interviews, revision of herbarium specimens, and bibliography. The quoted applications were: to treat pounds and wounds. For these purposes, the drug (leaf and stem) is prepared and administered as infusion, or as poultice. In order to provide data for inclusion of this species in the Herbal Pharmacopoeia of the United Mexican States (FHEUM) we describe its leaf architecture and diagnostic anatomical characters of drug,. The anatomical study revealed that the three types of trichomes observed can aid to differentiate this species from other Cuphea species. Anatomical variation was observed mainly in the midrib of the leaf. Leaf architecture is first described for this species, and provides the basis for future comparisons. In addition, the acetone-water extract of the aerial parts previously reported as cytotoxic activity against DU-145 (human prostate carcinoma) and HCT-15 (human colon carcinoma), showed the presence of polyphenolic compounds of the type of hydrolysable and condensed tannins, mannitol was isolated and identified.


Cuphea aquipetala Cav., Lythraceae (Hierba del cáncer) es utilizada en la medicina tradicional mexicana. En el presente trabajo se determinó mediante entrevistas, consultas de herbario y revisión bibliográfica sus aplicaciones médicas más frecuentes, a saber: para tratar golpes y heridas, para lo cual la droga (hoja y tallo) es preparada en infusión, o administrada en emplastos. Con la finalidad de aportar datos que ayuden a la inclusión de esta especie en la Farmacopea Herbolaria de los Estados Unidos Mexicanos (FHEUM), se describen la arquitectura foliar y los caracteres anatómicos de la droga. El estudio anatómico reveló que los tres tipos de tricomas reconocidos en la especie estudiada apoyan a diferenciarla de otras especies de Cuphea sp. Se observó variación anatómica principalmente en la nervadura central de la hoja. La arquitectura foliar se describe por primera vez para la especie y sienta las bases para futuras comparaciones. Por otra parte, el extracto acetona-agua de las partes aéreas que previamente ha sido reportado con actividad citotóxica contra células DU-145 (carcinoma de próstata humano) y HCT-15 (carcinoma de colon humano) mostró la presencia de compuestos polifenólicos del tipo de los taninos hidrolizables, taninos condensados, y se aisló manitol.

ABSTRACT

The "chía", Salvia hispanica L. (Lamiaceae), is an herbaceous annual plant native to Mexico. Its fruits contain antioxidant substances important for human nutrition, such as linoleic and alpha-linolenic acids, natural sources of omega-3 and omega-6 acids. The aim of this work was to establish morphological and anatomical diagnostic characters for leaf, stem, fruit and seed of the species in order to contribute to the botanical quality control in the marketing process as raw material for food and/or pharmaceutical industry.


Salvia hispanica L., "chía", (Lamiaceae), es una planta herbácea, anual, originaria de México. Sus frutos contienen antioxidantes, ácido linoleico y alfa-linolénico, fuente natural de ácidos grasos omega-6 y omega-3, importantes en la nutrición humana. El objetivo de éste trabajo fue establecer caracteres morfológicos y anatómicos diagnósticos de hoja, tallo, fruto y semilla de dicha especie, a fin de contribuir al control de calidad botánico en el proceso de comercialización como materia prima para la industria alimenticia y/o farmacéutica.

Quassia amara L. popularly known as “quasia”, is a shrubby plant from Tropical America. The wood, bark and leave are used in either folk medicine or in procuring phytotherapeutic drugs. The aim of the present work was to analize morphoanatomical and micrographic features which might provide assistance in the identification, analysis and standardization of Quasia amara L wood, bark and leaves. Results. Anatomical study showed white yellowish and diffuse porous wood, confluent paratracheal parenchyma. Rays width 1 cell wide and 8-30 cells high. CaOx crystals are absent. Cortex, 1-4 mm thick, a periderm up to 12 layers phellem cells. Leaf, hipostomatic with dorsiventral mesophyll and high number of sclerosed idioblasts.


Quassia amara L. popularmente conocida como “quasia” es un planta arbustiva de América Tropical. El leño, corteza y hojas son usadas tanto en medicina popular como en la obtención de drogas fitoterapéuticas. El objetivo del presente trabajo es analizar características morfoanatómicas y micrográficas las cuales provean asistencia en la identificación, análisis y estandarización de la madera, corteza y hojas de Quassia amara L. Resultados. El estudio anatómico mostró leño, blanco amarillento, de porosidad difusa. Parénquima paratraqueal confluente. Radios de 1 célula de ancho y 8-30 hileras de alto. Faltan cristales CaOx. Corteza, 1-4 mm de espesor, una peridermis de hasta 12 estratos de células de súber. Hoja, hipoestomática, con mesófilo dorsiventral, con elevado número de idioblastos esclerosados.

ABSTRACT

Endophytic fungi inhabit vegetable tissues or organs, without causing them any harm. Endophytes can co-evolve with plant hosts and possess species-specific interactions. They can protect the plant from insect attacks and diseases, and are also able to produce substances of biotechnological interest. In folk medicine, the bark, roots and fruits of Sapindus saponaria is used to produce substances with anxiolytic, astringent, diuretic and expectorant properties, as well as tonics, blood depuratives and cough medicine. This study evaluated the diversity of endophytic fungi present in the leaves of S. saponaria L. and observed the colonization of host plants by endophytes, using light and scanning electron microscopy. We verified that these fungi are found in intercellular and intracellular spaces. The genera of some isolates of S. saponaria were identified mainly by sequencing of ITS region of rDNA and, when possible, also by their microscopic features, as follows: Cochliobolus, Alternaria, Curvularia, Phomopsis, Diaporthe and Phoma. Phylogenetic analysis showed the existence of genetic variability of the genera Phomopsis and Diaporthe and interspecific variation among the Curvularia, Alternaria and Phoma, belonging to family Pleosporaceae.

ABSTRACT

Lantana montevidensis [Spreng.] Briq. is a member of family Verbenaceae, that includes about 98 genera and more than 2600 species which are predominantly tropical or subtropical plants. The leaf infusions of the plant are used in folk medicine to treat fever, influenza, asthma, bronchitis, and many other diseases. Biological studies showed that the plant has antimicrobial and antiproliferative activities. No detailed information could be traced concerning the macroand micromorphology of the plant. This motivated the authors to carry out this study to identify the drug in both entire and powdered form

ABSTRACT

Leaf epidermis ultrastructure of Zeugites (Poaceae: Panicoideae). The genus Zeugites comprises eleven species of neotropical grasses and it is principally distributed in Mexico, with some species extending to the Caribbean region, Central and South America. In this work, leaf epidermis ultrastructure of 11 species is described by the use of scanning electron microscopy. At least three specimens per species, that included herbarium and collected specimens, were used. An identification key and specific descriptions are included, in which the distinctive epidermal features are highlighted. The taxonomic valuable characters found were the following: presence or absence of prickles and macrohairs, intercostals short cells form and silica body form. Based on leaf epidermis characteristics, Zeugites species can be arranged into three groups: (1) species that lack prickles (Z. americana, Z. mexicana, Z. pringlei, Z. munroana and Z. sagittata); and lack macro hairs, with the exception of Z. pringlei; (2) species that have prickles (Z. latifolia and Z. smilacifolia); (3) species that have both, prickles and macrohairs (Z. capillaris, Z. hackelii, Z. pittieri and Z. sylvatica). El género Zeugites incluye once especies de pastos con distribución neotropical, en México, el Caribe, Centro y Sudamérica. En este trabajo se describe la epidermis foliar de las once especies, se utilizaron técnicas de microscopía electrónica de barrido y revisaron al menos tres especímenes de cada especie, algunas muestras proceden de ejemplares de herbario y otras fueron recolectadas por la primera autora. Se presenta una clave y descripciones para las especies del género, en las cuales se resaltan las características epidérmicas distintivas. Los caracteres que resultaron informativos, desde el punto de vista taxonómico fueron: la presencia o ausencia de aguijones y macropelos, la forma de las células cortas intercostales y la forma de los cuerpos de sílice. Con base en estas características, las especies de Zeugites se pueden separar en tres grupos: (1) especies que no presentan aguijones (Z. americana, Z. mexicana, Z. pringlei, Z. munroana y Z. sagittata) y que con excepción de Z. pringlei tampoco presentan macropelos; (2) especies que presentan sólo aguijones (Z. latifolia y Z. smilacifolia); y (3) especies que presentan aguijones y macropelos (Z. capillaris, Z. hackelii, Z pittieri y Z. sylvatica). Las características morfológicas de la epidermis foliar, sustentan la relación de las tribus Centotheceae y Paniceae.

Plant biominerals are not always well characterized, although this information is important for plant physiology and can be useful for taxonomic purposes. In this work, fresh plant material of seven wild neotropical species of genus Canna, C. ascendens, C. coccinea, C. indica, C. glauca, C. plurituberosa, C. variegatifolia and C. fuchsina sp. ined., taken from different habitats, were studied to characterize the biominerals in their internal tissues. For the first time, samples from primary and secondary veins of leaves were investigated by means of infrared spectroscopy, complemented with X-ray powder diffractometry and scanning electron microscopy. The spectroscopic results, supported by X-ray powder diffractometry, suggest that the calcium oxalate is present in the form of whewellite (CaC2O4×H2O) in all the investigated samples. It is interesting to emphasize that all IR spectra obtained were strongly similar in all species studied, thus indicating an identical chemical composition in terms of the biominerals found. In this sense, the results suggest that the species of Canna show similar ability to produce biogenic silica and produce an identical type of calcium oxalate within their tissues. Los biominerales de las plantas no siempre han sido bien caracterizados aunque esta información es importante en fisiología vegetal y puede ser de utilidad para fines taxonómicos. En este trabajo se estudió material vegetal fresco de siete especies silvestres neotropicales: Canna, C. ascendens, C. coccinea, C. indica, C. glauca, C. plurituberosa, C. variegatifolia and C. fuchsina sp. ined., provenientes de diferentes localidades, con el fin de caracterizar los biominerales presentes en sus tejidos foliares internos. Por vez primera, muestras de venas primarias (ejes foliares) y secundarias de hojas de estas especies se investigaron por medio de espectroscopia de infarrojo, complementada con estudios por difracción de rayos X de polvos y microscopía electrónica de barrido. Los resultados indicaron la presencia de ópalo (sílice biogénica) y oxalato de calcio en los tejidos vegetales analizados. Además, se determinó que el oxalato de calcio está presente en forma de whewellita (CaC2O4×H2O), información nueva para el género. Tanto el ópalo como la whewellita están presentes en todas las especies analizadas, que representan aproximadamente un tercio de las especies silvestres del género. La capacidad de biomineralizar SiO2 en forma de ópalo en especies de Canna de diversos ambientes resulta también un rasgo altamente sugerente para futuros estudios.

ABSTRACT

The micromorphology of foliar trichomes of Hypoestes aristata var. aristata was studied using stereo, light and scanning microscopy (SEM). This genus belongs to the advanced angiosperm family Acanthaceae, for which few micromorphological leaf studies exist. Results revealed both glandular and non-glandular trichomes, the latter being more abundant on leaf veins, particularly on the abaxial surface of very young leaves. With leaf maturity, the density of non-glandular trichomes decreased. Glandular trichomes were rare and of two types: long-stalked capitate and globose-like peltate trichomes. Capitate trichomes were observed only on the abaxial leaf surface, while peltate trichomes were distributed on both adaxial and abaxial leaf surfaces.

ABSTRACT

Simira is a predominantly woody Neotropical genus comprising 41 taxa, 16 of which occur in Brazil and eight of them in the southeastern region of Brazil. Leaf blades of Simira eliezeriana Peixoto, S. glaziovii (K. Schum.) Steyem., S. grazielae Peixoto, S. pikia (K. Schum.) Steyerm., S. rubra (Mart.) Steyerm., S. sampaioana (Standl.) Steyerm. were collected in the southeastern region of Brazil and fixed according to usual methods for light and electron microscopy. The leaf blades show typical characteristics of the Rubiaceae family as dorsiventral mesophyll and paracytic stomata. The presence of two bundle sheaths that extend to the upper epidermal layer, prismatic crystal and crystal-sand, alkaloids in the mesophyll and the organization micromorphological of the outer periclinal wall are considered characteristics representative for the genus. This study also demonstrates some leaf blade characteristics that can be used to Simira species identification (leaf surface, domatia types, epicuticular wax types and patterns of epidermis anticlinal cell walls).

ABSTRACT

Pharmacognostic investigation of the leaf of Tapinanthus bangwensis [Engl. and K. Krause] Danser was carried out using phytochemical and anatomical methods. Polygonal epidermal cell shape, straight - curved anticlinal wall pattern, paracytic stomata and presence of the crystals of calcium oxalate in the cell lumen are characteristic of the species. Other useful identification indices include glabrous leaf, arc-like closely spaced vascular bundle in the petiole and centrally located vasculature of the midrib. The phytochemical characters like tannins, saponins, cardiac glycosides, reducing compounds and flavonoids are present but anthraquinones, alkaloids, phlobatannins and cyanogenetic glycosides are lacking. With these features, the plant can be identified without ambiguity and the chemical constituents underlie the potency of the plant as a plant of high medicinal value used in traditional medical practice for inhibiting the growth of bacteria, fungi and virus, and also in rectifying sexuality problems

ABSTRACT

Oryza grandiglumis is a wild species of rice endemic to tropical America. This species was first found in 1998 in the wetlands of Caño Negro, located in the northern part of Costa Rica. Twenty five plants of O. grandiglumis were processed for scanning electron microscope. An ultrastructural description of the leaf blade, ligule, auricles, spikelet and caryopsis, with an emphasis on structures of taxonomic value. The leaf blade has a characteristic cuticular wax pattern, composed of dense rod-like structures, and is surrounded by papillae, zipper-like silica cells, abundant bulky prickle trichomes, and hooked trichomes. The blade's edge has three rows of hooked prickle trichomes of various sizes. The auricles wrapped the culm, with long attenuated trichomes at the edges; the base was surrounded by oblong cells. The ligule is a blunt membrane covered by short prickle trichomes. Spikelet morphology is characteristic of the Poaceae family, but the sterile lemmas were nearly as long as the fertile lemmas, and they have an unique crown-like structure of lignified spines between the rachilla and the fertile lemmas. Comparison with Brazilian specimens of O. grandiglumis revealed little differences in the ultrastructural characteristics.


El arroz silvestre Oryza grandiglumis es endémico de América. Se localiza en la zona norte de Costa Rica, principalmente en el humedal de Caño Negro y del Río Medio Queso. Es una planta vigorosa y grande. Su nombre deriva del gran tamaño de las lemas estériles (glumas). Presentamos una descripción ultraestructural de la lámina foliar, lígula, aurículas, espiguilla y cariópside, con énfasis en las estructuras de valor taxonómico, usando el microscopio electrónico de barrido. La lámina foliar se caracteriza por presentar un patrón de cera cuticular en forma de densos bastoncillos. Presenta estomas rodeados de papilas, células de sílice en forma crenada, varias formas de papilas de cera, distribuidas en forma muy regular, y tricomas espinosos abultados en la base y tricomas ganchudos. En el borde de la lámina hay tres hileras de tricomas espinosos ganchudos de diferente tamaño. Las aurículas son envolventes y los bordes presentan tricomas atenuados largos cubiertos en la base por células alargadas. La lígula es truncada y cubierta de tricomas espinosos pequeños. La morfología de la espiguilla es algo similar a las de las otras especies de la familia Poaceae, pero las lemas estériles son casi del mismo tamaño que las fértiles. Además se observó entre la raquilla y las lemas fértiles una corona de espinas lignificadas. Ultraestructuralmente, esta población es similar a las brasileñas.
